Analysis of Association Between the Effects of Methylphenidate and DRD4 Gene Polymorphisms in Patients with Attention Deficit Hyperactivity Disorder

Background: Drug treatment is one of the most important treatments for attention deficit hyperactivity disorder (ADHD). The DRD4 gene a transporter and receptor coding dopamine genes under investigation in etiology ADHD. In this study, association between rs3758653 C/T VNTR exon 3 repetition polymorphisms effects methylphenidate were investigated patients with ADHD consuming methylphenidate. Methods: descriptive-analytical study was performed on 122 (5 - 18 years old) who treated DNA extracted using salting out method. Subsequently, polymorphism 5'UTR region genotyped by PCR-RFLP method, fragment III electrophoresis gel acrylamide After eight weeks from start drug methylphenidate, intensity symptoms evaluated Conners scale. Finally, all data questionnaires information that resulted laboratory findings analyzed ANOVA repeated measure analysis. Results: Of 15 (12.3%) responded to treatment, 107 (87.7%) not responded. significant differences revealed genotype, allele frequencies (C/T) 3'VNTR repeats responder non-responder groups treatment. Conclusions: results showed reduction related sub-types
